Ph.D. Thesis Title

Study of Restructuring Decisions and Complexity of Lean Manufacturing Systems  

The fundamental contribution of the thesis is the development of new methods of analysis for more effective quantitative comparison and analysis of restructuring decisions in manufacturing systems. The new methods are proposed based on the physical interpretation of the existing quantitative methods of coefficients of dissimilarity based on the results of graph theoretic models in a multi-dimensional Euclidian space and have a focus on complexity reduction in lean manufacturing systems. The study investigated the case of work flow restructuring in a steel plant and two cases of work flow restructuring in a packaging equipment industry for implementing lean philosophy and it used graph theoretic modeling for analysis. The real performance indicators such as improvement in on-time delivery performance from industrial case studies indicate a strong correlation with the values of proposed indices of complexity based on graph theoretic modeling.

Brief about the company 

Tube Investments of India is a flagship company of multi billion dollar conglomerate of Murugappa Group. It is a market leader in the area high quality cold drawn welded (CDW) and electric resistance welded (ERW) tube products.

 

Main Contributions/Achievements (Link to Poster)

Through this project, three major alternative lubrication systems have been investigated for tube drawing process namely

  • the use of tin based porous layer on tube surface to better retain lubricant, 
  • the use of solid tin layer as lubricant during tube drawing process
  • the use of ultrasonic vibration for die and plug during tube drawing process  

Improvements in the surface finish to the order of 40 percent have been achieved through these investigations.

Research Interest

The laboratory is dedicated to the structural and molecular understanding of protein folding and stability, protein-protein interactions and structure-function relationship in proteins. The ubiquitous nature of a new class of hemoglobins in plants, the presence of novel hemoglobins in the microbial world, and the discovery of new hemoglobins in humans and other vertebrates have bolstered the hemoglobin research over the last several years. A major focus of the laboratory is the investigation of novel algal, plant, bacterial and animal hemoglobins involving techniques like laser flash photolysis, stopped-flow spectrometry, FTIR and EPR spectroscopy, site directed mutagenesis, computational biology and X-ray crystallography. Emphasis is on deciphering the mechanism of regulation of ligand binding in novel (hexacoordinate and truncated) hemoglobins in general. The goal is to be a part of the effort to set up a ‘biophysical fingerprint’ for novel hemoglobins that can help assign physiological functions. Other related proteins like hemoglobin reductases are being simultaneously investigated for a better understanding of function(s) of the novel globins. Stability, aggregation and protein folding problem related to classical and novel hemoglobins as well as hemoglobin based artificial blood substitutes is being simultaneously investigated. Comprehensive knowledge accumulated by hemoglobin researchers world-wide may be useful in improving oxygen transport and storage in mammalian circulatory systems, nitrogen fixing efficiency in plants, sensing and response to hypoxic conditions, scavenging efficiency under stress, and our ability to use heme protein based artificial oxygen carriers. The latter is being fervently pursued in the laboratory because of its translational value in medicine. The laboratory is also focused on spectroscopic and mass spectrometric characterization/screening of hemoglobin disorders with a goal to set up a simple, fast, economic and reliable diagnostics for such disorders using very small amounts of samples. The group is further engaged in understanding the pathophysiology of sickle cell disease using metabolomics and proteomics approaches and is attempting to decipher effective therapeutics to combat the debilitating disease, with promising success.
 
Structure-based rational drug discovery and design is currently the major emphasis of the laboratory. The aim is to seek structural insight of human dopamine b-hydroxylase (DBH) and ADP Ribosylation Factor Like Protein 15 (ARL15), drug targets for complex traits. Availability of the three-dimensional structures of these proteins will help detailed investigation of the structure-function relationship in these important enzymes and lay a platform for rational therapeutic drug design. The laboratory is currently focused on the discovery of potential small molecule inhibitors of DBH and ARL15 to combat cardiovascular diseases and rheumatoid arthritis, and a few lead molecules have emerged that might be developed as drugs for the target non-communicable diseases. On the other hand, there are attempts to identify small molecules to combat infectious diseases like leishmaniasis, as well as cancer.

   A Reusable Machine-Constructed Feature Representation for Protein Sequences

 

Multi Purpose Protein Prediction Vector (MP3vec) representation is constructed from a protein’s sequence and its PSSM profile using all the available high resolution protein structural data with the aim to aid in 'transfer learning' to overcome the 'small data' problem in developing predictive models for biomacromolecular interactions. Please refer to the Readme file and the binary can be downloaded as a zip file.

Molecule Property Predictor

 

With the advancement in Artificial Intelligence, it has been possible to predict things helpful in
chemistry and biology, including drug discovery. Supervised learning has been very fruitful in
predicting the physicochemical properties of small molecules. These models are trained using a
labeled dataset, where the model learns about each data type. The dataset here has 133k stable
small organic molecules with nine heavy atoms (CONF) out of the GDB-17 chemical universe.
A message-passing neural network (MPNN) has been discussed in detail in this paper, along with
novel variations to increase the accuracy of the supervised learning models. The MPNN works
on the idea of the message, update, and readout functions that operate on different graph nodes.
These variations include revamping the input system and adding a semi-master node in the
architecture to increase the overall accuracy to create a robust model. Improvements in the model
will lead to a scalable system for choosing compounds to improve and discover the drugs.

Aim: The main objective of this project is to develop integrated hardware for in-situ monitoring and control of solar panels. Ongoing prototype testing and pilot implementation in Hyderabad

Status: completed

Findings and summary: Panel level monitoring hardware was developed to understand and address the anomalies of power generation in a solar array at 2 sites of ATUM in Hyderabad. ATUM manufactures Building Integrated Photovoltaic Modules. PPV  developed an integrated software and hardware solution for them to test their installations’ efficiency under partial shading and soiling conditions and improved overall performance by 2-2.5%

Research Interest

Role of alternativesplicing in metabolic disorders: Alternative splicing is a major source of diversity in the human proteome, which can modulate protein function and affinities for the assembly of heteromeric complexes & also alter mRNA stability. 

 Evidence on the roles of splicing and alternative splicing in various diseased conditions is emerging. However, the role of alternative splicing linking signaling pathways to gene regulation and the onset of various metabolic syndromes, for example, type II diabetes, is not well understood. It is important to know the genes involved in these diseases and their mode of regulation. Knowledge at the molecular level would thus provide the future foundation of research and drug discovery.

The objective of the current study is to identify genes that undergo changes in alternate splicing upon receiving different stimuli that contribute towards metabolic syndromes like obesity, insulin resistance, and type II diabetes and study their molecular mechanism(s) of actions. This study will help link signal transduction and gene regulation by alternative splicing and diseased conditions and will thus be important for clinical research and drug discovery.

Role of alternative splicing in stem cell differentiation:This study is aimed to investigate whether alternative splicing, one of the major sources of gene regulation and protein diversity, plays a role in the process of differentiation of pluripotent and multipotent stem cells. Manipulating the expression of one or more splicing regulators may accelerate the rate of differentiation into specific lineages, which is a major goal in regenerative medicine.

Targetedgene delivery as potential therapeutics: The main aim of this collaborative work is to manipulate gene expression profiles as potential therapeutics by long-term silencing/overexpression of genes of interest using biocompatible nano-vehicles for gene delivery.

We are developing an all-water-based strategy to create flexible electronics, sensors, and drug-delivery devices for physiological applications. This work is in collaboration with Prof. Srinivasa K Prasad from the Department of Pharmacy @BPHC.
Cold Plasma-based Advanced Oxidation Process

We are studying the effect of cold plasma on the oxidation of antibiotic residues in water. This work is in collaboration with Prof. G. R. Abhijith from the Department of Civil Engineering at IIT Kanpur and Dr. Neethu Bhasker from  KSCSTE.

Non-enzymatic Glutamate Sensors

In this project, we are developing copper nanoparticle-based glutamate sensors. 


Development of Bacteria Detection Sensors

In collaboration with Prof. Debirupa Mitra, we are trying to develop point-of-care diagnostic tools for detecting infection in wounds.

Shape-memory polymers

We are currently studying the effect of water uptake on the actuation performance of shape memory polymers. This work is going on in collaboration with Prof. Punnag Chatterjee from the Department of Mechincal Engineering at IIT Dharwad.

As a Research Associate

  • This project, sponsored by Council for Scientific and Industrial Research (CSIR), focused on developing an experimental facility for multistage liquid – sold fluidized bed adsorption for the removal of pollutants from wastewater. Experimental and theoretical studies were carried out for understanding the said system for the removal of colour from wastewater.

As a Senior Project Officer

  • I worked on a DNES, Department of Non Conventional Energy Sources, Ministery of Energy (MoE), sponsored project on “Generation of Biogas from Industrial Wastewater – Tannery Waste”. Three contact filters were designed and constructed. Studies were focused on anaerobic degradation / treatment of Tannery wastewater with the main aim of biogas – generation.

Details: Develop a mathematical model by estimating the exact position of the nodes in the network in a particular time interval. Design algorithms for maintaining the connectivity and density estimation of the nodes. Test and validate the results obtained by the proposed strategy.
 
Methodology: The methodology for providing the solution to the problem of reliable data
dissemination consists of defining a prototype as per the requirement of users needs, e.g.,
the prototype consists of the mathematical formulation of finding the location and mobility of the vehicles in a particular region during different time intervals. Then various algorithms
would be proposed to solve the problem of connectivity and density of the nodes. The algorithm controls the communication among the different nodes in the network in a collaborative manner, i.e, various nodes in the network share the information such as routing decisions, mobility patterns etc. with each other so that necessary steps can be taken for smoother action in the network. Finally, the designed algorithms would be tested analytically and by simulation with respect to various parameters in comparison to the existing solutions in the literature.
 
The scope of the proposal: As VANETs is an emerging technology so it can be used in wide
variety of applications such as construction of ITS, home security, surveillance system, alert
generation in case of emergency in the community or on the roadside etc. All these applications require an efficient data dissemination approach along with the reliability. So the current proposal will try to solve the problems in the existing proposals such as long end to end delay in dense regions and poor connectivity in a particular geographical region.

Objectives: The project works with following objectives: I. Reduce the power consumption of network without impacting the quality of service. We would design our solution in such a manner that the QoS that is promised to the client will not be impacted. For that, we would take care of load balancing, extra backup paths, prioritization to those flow which is of higher priority. II. Designing a new algorithm and controller core that will assist in taking decision in real time based on quick switching between device state. Apart from an algorithm which will be based on previous history and current network condition for better performance and strategic decision, for quick switching we will implement a finite state machine with proper input/output and action condition. III. Building efficient data structure for collecting the statistic and getting useful insight without taking much time. We would focus our energy to build a data structure which will take minimum time complexity for storing and retrieving the information. Information will be stored in form of codecs so that we will also minimize the space complexity. The new data structure will be either a derivative of hash that takes constant time for action or based on the tree that takes logarithmic time for taking any decision.

Mobile App for assisting Indian farmers in improving yield by automatic identification of plant diseases (Rice and Tomato) [  Funded by DST SYST, Ongoing]  as PI.

India suffers from annual crop loss of many crores of rupees owing to pest and disease infestation, according to a study by Assocham and Yes bank. Different plants are infested by different diseases; while some are caused by fungi, some are caused by bacteria and some by viruses.  This project aims in automatically identifying key diseases in specific plants using mobile application, so that the farmers can be benefitted. The image database pertaining to the Indian subcontinent, would be employed to train a model that can automatically predict plant diseases. Both traditional image processing techniques as well as the recent advances in convolutional neural networks would be employed for this purpose.

Brief Description of the Project: The proposed research project intends to define and explore the Chinese conceptualization of the Harmonious World and its characteristics as a theoretical framework to comprehend China’s bilateral engagement with countries like Sri Lanka. The purpose is to understand the nature of the Harmonious World and investigate whether the Chinese concept of a Harmonious World attempts to redefine/challenge the normative underpinnings of sovereignty, paving its way for a hierarchical or Sino-centric global order and, if yes, how. Accordingly, this study examines China- Sri Lanka engagement in the post-civil war scenario, and most of this engagement has been accredited as the Belt and Road Initiative (BRI).
